Protein engineering is a widespread approach in the branch of protein science. The elucidating function of a single or a stretch of amino acids is a useful tool for Protein engineering. The improvement in the properties of a protein is used by some protein engineers. The art or science of making practical applications of the knowledge of pure sciences is Protein Engineering. It is one of the most exciting developments in molecular biology has been the ability in the construction and design of new proteins or enzymes with novel functions. Recombinant DNA technology, is possible to modify the amino acid sequence of a protein, including substitution, deletion, or insertion of amino acid residues or to fuse two or more proteins to form hybrid proteins, or to totally synthesize genes for new proteins. The Focus areas include optimization of protein performance and production using techniques and knowledge from protein design and optimization, protein purification, quality control, crystal structure determination, 3D structure prediction, assay design, enzyme specificity and kinetics, physical properties and complex enzyme reactions. It has generated enzyme variants with improved catalytic activity, broadened or altered substrate specificity, as well as raised or reversed stereo selectivity. To retain activity at elevated temperatures and to function in the presence of organic solvents, salts and pH values far from physiological conditions these enzymes have been tailored.
